چکیده

Mammalian phosphoinositide-specific phospholipases C (PI-PLCs) are involved in most receptor-mediated signal transduction pathways. The mammalian isozymes employ a modular arrangement of domains to achieve a regulated production of two key second messengers. The roles of the PH, EF hand, C2, SH2 and SH3 modules in regulation of these enzymes and in interactions with membranes and other proteins is becoming apparent from recent structural and functional studies. ß 1999 Elsevier Science B.V. Phosphoinositide-3-Kinase Is the Primary Mediator of Phosphoinositide-Dependent Inhibition in Mammalian Olfactory Receptor Neurons

Odorants inhibit as well as excite primary olfactory receptor neurons (ORNs) in many animal species. Growing evidence suggests that inhibition of mammalian ORNs is mediated by phosphoinositide (PI) signaling through activation of phosphoinositide 3-kinase (PI3K), and that canonical adenylyl cyclase III signaling and PI3K signaling interact to provide the basis for ligand-induced selective signa...

D5 dopamine receptors are required for dopaminergic activation of phospholipase C.

Dopamine activates phospholipase C in discrete regions of the mammalian brain, and this action is believed to be mediated through a D(1)-like receptor. Although multiple lines of evidence exclude a role for the D(1) subtype of D(1)-like receptors in the phosphoinositide response, the D(5) subtype has not been similarly examined. Here, mice lacking D(5) dopamine receptors were tested for dopamin...

Multiple forms of phosphoinositide-specific phospholipase C and different modes of activation.

Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in initiating the surface-mediated signal transduction by generating second messenger molecules, diacylglycerol, and inositol phosphates (see [ 1-41 for reviews). Results accumulated during the last few years clearly indicate that the response of phosphoinositide signalling to a specific stimulus varies depending on tissue and...
